I'll put in another vote for the Marx Brothers. Besides the obvious choices--Duck Soup and A Night at the Opera--Horse Feathers is worthy of mention for what is perhaps the funniest cinematic football game this side of MASH.

Monty Python and the Holy Grail (of course)
Real Genius
To Wong Foo Thanks For Everything Julie Newmar (because Wesley Snipes in stilettos gets me every time)
A Fish Called Wanda
and
Sixteen Candles

it's too difficult to pick the funniest movie. but my most favorite scene in a movie is in serial mom when kathleen turner's character is prank calling her neighbor, hehe .

Default Meet the Feebles

Here's some right off the top of my head.

South Park: Bigger Longer & Uncut
The Big Lebowski
Dirty Rotten Scoundrels
Three Amigos
Flying High (aka Airplane)
Naked Gun trilogy
Austin Powers trilogy

But my personal favourite has to be Meet the Feebles. Before he became Lord of the Multiplex, Peter Jackson wrote and directed this all time classic about a demented puppet variety show.
It stacks so much into its narrative - love, gluttony, revenge, sex, drugs, pornography, songs, a junkie snake who suffers 'Nam flashbacks, walruses playing golf, a gun toting hippo, ... the list goes on & on It also pokes some spot-on satirical stabs at the entertainment industry and the press. It's been described as "the Muppets on Acid" and I can think of no better compliment than that. It's totally insane, extremely gross, but if you appreciate lowbrow humour, Meet the Feebles has a constant stream of LOL moments. It may be hard to find in the video store, but it's worth the search. Come to think of it, Feebles could have quite feasibly been a source of inspiration for Matt & Trey ...
